Yes !!! the same PCR program may have slightly different completion time for the same reaction (having the same number of cycles) in two different thermo-cyclers due to difference in the ramp rate of the machines. For example if I keep two sets of PCR with the same program; one in a Bio-Rad machine and the other one in an Eppendorf machine then one may have a completion time of 1hr 48mins and the other may have completion time of 2hrs 5mins.
So many questions bothering you; but you don't have time to find the answer nor anybody in your lab has time to answer it or sometimes only one little thing is bothering you every-time everywhere!!! There are people those have faced the same problem during their work but now are with solutions.All you need to do is just ask!!! Yes. It depends on the ramp rate of machine. Ramp rate is the time required for heating/cooling the block. More the ramp rate, less the reaction time required. Earlier Eppendorf machines were of 3 ramp rate (temperature rises 3 degrees for every seconds). But now many machines are with ramp rate 5 or 6 also. This reduced the reaction time drastically.
